Police were called to a Thomastown home about 8.40pm on Thursday and found an unconscious 36-year-old Doreen man with stab wounds.
He died at the property, on Victoria Drive, despite efforts to resuscitate him.
A 45-year-old Glenroy man was arrested at the scene, while detectives are searching for a second person.
"At this early stage in the investigation, the incident appears to be a targeted attack," a police spokeswoman said.
Police want to speak to anyone with information about the stabbing, or those in the area who may have CCTV or dash-cam footage.
