Her guest-starring character Yasmine "Yas" Shields joins the show as a "glamorous and successful businesswoman in pursuit of an exciting new opportunity" when filming starts in July.
"I am honoured and excited to join such a beloved and iconic show," Ms Stause said in a statement.
"Coming from the world of soaps in the U.S., it's back to my first love in the entertainment industry," she said.
"They pitched me an idea for a character, and I was immediately excited to figure out a way to make it happen. Ramsay Street here I come!"
Soap operas All My Children and Days of Our Lives gave Ms Stause her start on television before she retrained as a real estate agent and landed a role flipping Los Angeles mansions on Netflix's Selling Sunsets.
In 2022 she became an honourary Aussie by marrying Melbourne-born singer, songwriter and multi-instrumentalist G-Flip.
Ms Stause was the second high-profile US actor to join Toadfish Rebecchi in Erinsborough after former star of The O.C. Mischa Barton was added to the cast in 2023.
Neighbours executive producer Jason Herbison said the cast and crew were "thrilled" to welcome Ms Stause to the show.
"We created a character especially for her and we can't wait for her to bring the storyline alive," he said.
"Expect intrigue and surprises - and many implications for the residents of Ramsay Street."
Neighbours returned to the airwaves after a brief cancellation in 2022. The show, which started in 1985, was rebooted by Amazon Freevee in 2023 and has since been nominated for two Daytime Emmys.
