The closures come into effect from Thursday in addition to authorities shutting the Twelve Apostles and Port Campbell visitor centres about three weeks ago.
A spokesman said the closure of eight highly visited sites in western Victoria would remain "until further notice" to slow the spread of coronavirus.
"This Easter the message is clear. Stay home. Protect our health system. Save lives," a statement said.
Other parks and reserves remain accessible for exercise if physical distancing requirements are strictly followed.
Parks Victoria rangers will patrol open and closed parks, working with Victoria Police and authorised officers from the Department of Environment, Land, Water and Planning to monitor compliance with the Chief Health Officer's directions.
Victoria Police can issue on the spot fines of up to $1652 for individuals and up to $9913 for businesses who don't follow the directions.
Parks Victoria says it will continue to monitor visitation rates with the possibility of further changes to park closures.
The additional parks and reserves closed in Western Victoria include:
- Bay of Islands Coastal Park
- Discovery Bay Coastal Park
- Grampians National Park
- Great Otway National Park (partially open for local residents to exercise only)
- Lower Glenelg National Park
- Mount Arapiles-Tooan State Park (partially open for local residents to exercise only)
- Port Campbell National Park
- Tower Hill Wildlife Reserve
