North Warrnambool Warriors drew with Horsham Panthers in the final round on Saturday - the best result of its inaugural campaign.
Warrnambool Raiders and Gunditjmara Bulls will vie for the premiership after securing their spots in the top five.
The Raiders finished atop the ladder, earning week one of the three-week finals series off.
The Bulls, who finished second, will play third-placed Naracoorte Jets in a semi-final on Saturday.
Blue Lake Knights and Horsham Panthers will face-off in an elimination final.
Gunditjmara Bulls coach Renato Da Ros said his team, which finished the home-and-away season with a 50-20 drubbing of Blue Lake, was wary of the Jets.
"We'll have to pick up our game against the Jets if we want to beat them," he said.
"We have to concentrate on defence and our tackling has to be pretty well spot on.
"We'll take the game to them and apply as much pressure as we can."

North Warrnambool Warriors player Tyne Anderson said the 2019 season was about developing a club culture.
"The way we wanted to run the club in the first year was as a family club and that's exactly the way it has turned out," he said.
Anderson said the Warriors had also built a strong foundation with Purnim Cricket Club which shares the same venue.
"They let us go out and cook Thursday night meals to get money for the club and want us to continue to do it," he said.
Anderson played alongside brother-in-law Tahi Pompey, who returned from injury in the final round, at the Warriors.
"I play footy at Woorndoo-Mortlake and the deal was if Tahi played football I would play rugby," he said.
