Friends of the Gellibrand River, Estuary and Wetlands is organising the rally, which will include a march and opportunity to sign a petition.
Friends member Kim Morton said the group was not opposed to development, but believed the chosen site was inappropriate.
The group said the area was home to rare and migratory birds and an important fish breeding habitat. It fears the development and associated infrastructure could cause large-scale damage to the wetlands precinct.
The event will also include a wetland and estuary guest speaker and a chance to learn more about the area. It will be held at the Princetown Recreation Reserve from 11am Sunday.
Developer Gavin Ronan told The Standard earlier this month that extensive studies and planning had been undertaken to ensure the proposed resort struck the right balance between designing a successful eco-tourism venture and minimising its footprint.
